目的:研究地榆中升高白细胞的有效成分。方法:利用环磷酰胺致小鼠白细胞减少模型筛选地榆升高白细胞的有效部位,采用聚酰胺、硅胶、Sephadex LH-20和ODS C_(18)柱色谱对有效部位进行分离纯化,通过理化常数和波谱分析确定结构,并利用M-NFS-60细胞筛选地榆升高白细胞有效成分。结果:从有效组分中分离鉴定得到8个三萜类成分,分别为3β,19α-dihydroxyurs-12-en-28-oic acid 28-β-D-glucopyranoside(Ⅰ)、2α,3β,19β-trihydroxyurs-12-en-28-oic acid 28-β-D-galactopyranosyl ester(Ⅱ)、3β-[(α-L-Ara-binopyranosyl)oxy]-urs-12,18-dien-28-oic acid 28-β-D-glucopyranosyl ester(Ⅲ)、3β,19α-hydroxyolean-12-en-28-oic acid 28-β-D-glucopyranoside(Ⅳ)、kaji-ichigoside Fl(Ⅴ)、ziyu glycoside I(Ⅵ)、3β-[(α-L-Ara-binopyranosyl)oxy]-23-hydroxyurs-12,19(29)-dien-28-oic acid 28-β-D-glucopyranosyl ester(Ⅶ)、3β-[(α-L-Arabinopyranosyl)oxy]-urs-11,13(18)-dien-28-oic acid 28-β-D-glucopyranosyl ester(Ⅷ)。经细胞实验筛选,确定地榆乙酸乙酯部位中化合物I、化合物Ⅵ具有显著促进M-NFS-60细胞增殖的作用。结论:地榆乙酸乙酯部位具有最佳升高白细胞活性,其主要有效成分可能为化合物I和化合物Ⅵ。
Objective: To study the active ingredients of elevated leukocytes in Burnet. Methods: The effective sites of white blood cells were screened by cyclophosphamide-induced leukopenia model. The effective fractions were separated and purified by polyamide, silica gel, Sephadex LH-20 and ODS C 18 column chromatography. And spectral analysis to determine the structure, and the use of M-NFS-60 cells screened Ulmus leukocyte active ingredients. Results: Eight triterpenes were isolated and identified from the active components, which were 3β, 19α-dihydroxyurs-12-en-28-oic acid 28- trihydroxyurs-12-en-28-oic acid 28-β-D-galactopyranosyl ester (Ⅱ), 3β - [α-L-Ara-binopyranosyl) oxy] -urs- 12,18-dien-28-oic acid 28 -β-D-glucopyranosyl ester (Ⅲ), 3β, 19α-hydroxyolean-12-en-28-oic acid 28- Β-D-glucopyranosyl ester (Ⅶ), 3β - [(α-L-Ara-binopyranosyl) oxy] -23-hydroxyurs- -L-Arabinopyranosyl) oxy] -urs-11,13 (18) -dien-28-oic acid 28-β-D-glucopyranosyl ester (VIII). After screening by cell experiment, compound I and compound VI of ethyl acetate fraction of Ulmus pumila were determined to significantly promote the proliferation of M-NFS-60 cells. CONCLUSION: Ethyl acetate fraction of Ulmus pumila maximizes the activity of leukocyte. The main active components of compound E could be compound I and compound VI.
