目的探讨柴胡皂甙A(Ss A)对脓毒症大鼠早期血清中炎症因子的影响。方法 90只清洁级SD大鼠,随机分为对照组、脓毒症组和治疗组(脓毒症+Ss A),各30只。通过盲肠结扎穿孔建立脓毒症大鼠模型,术后1、8、24 h三个时间点收集动脉血标本。酶联免疫分析法检测血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)浓度。结果与对照组比较,脓毒症组大鼠血清TNF-α、IL-6水平在术后各时间点均明显升高(P<0.05)。在8、24 h时间点,治疗组大鼠TNF-α、白细胞介素-1(IL-1)的含量均较脓毒症组各时间点显著降低(P<0.05)。结论 Ss A可抑制脓毒症大鼠早期炎症反应。
Objective To investigate the effect of saikosaponin A (Ss A) on the early serum inflammatory cytokines in septic rats. Methods Ninety clean SD rats were randomly divided into control group, sepsis group and treatment group (sepsis + Ss A), each 30 rats. The model of sepsis was established by cecal ligation and perforation. Arterial blood samples were collected at 3, 8, and 24 hours after operation. Serum levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were detected by enzyme-linked immunosorbent assay. Results Compared with the control group, the levels of TNF-α and IL-6 in sepsis group were significantly increased at all time points (P <0.05). At 8 and 24 h, the levels of TNF-α and IL-1 in the treatment group were significantly lower than those in the sepsis group at each time point (P <0.05). Conclusion Ss A can inhibit the early inflammatory response in septic rats.
