本试验目的:用结实率较高的品种间杂种胚为试材,作胚培养条件的试验研究。试图掌握适宜于小麦胚的培养条件与方法,从而为大小麦杂种胚的人工培养奠定基础。材料和方法试材为73(36)-1-9-26×官村1号的 F_1 15天杂种胚作幼胚离体培养。基本培养基(mg/升):T.K 和 white 两种。附加成分:水解乳蛋白(CH)100—300,动力精(KT)1—2,吲哚乙酸(IAA)0.5—1.0。蔗糖浓度8%,琼脂0.8%,PH 值5.8。灭菌方法:幼胚先经70%乙醇表面灭菌2—3秒,转入0.1%氯化汞溶液浸泡5分钟,然后用无菌水冲洗3次。在无菌条件下将果实内幼胚剥离(或切割)下来,直接(不用褓姆法)
The purpose of this experiment: With higher seed setting rate of hybrid embryos as materials, embryo culture conditions of the experimental study. Sterilization: embryos first 70% ethanol surface sterilization 2-3 seconds, into 0.1% mercuric chloride solution for 5 minutes, then rinse with sterile water 3 times. Aseptically dissect (or cut) the immature embryos in the fruit directly (without using the Lim method)
