目的:在卵清蛋白致敏变应性鼻炎豚鼠模型基础上,采用糖皮质激素、羟基脲、腺嘌呤3种不同阳虚诱导剂诱发,建立变应性鼻炎肾阳虚复合模型,以筛选建立理想而稳定的变应性鼻炎(Allergic Rhinitis,AR)病证结合动物模型。方法:采用卵清蛋白(Ovalbumin,OVA)诱导建立变应性鼻炎豚鼠模型,同时给予糖皮质激素、羟基脲或腺嘌呤建立肾阳虚复合模型,观察动物宏观体征及死亡情况、局部过敏症状,酶联免疫吸附法(Enzyme-linked Immunosorbent Assay,ELISA)法测血清中s Ig E和组胺含量,苏木精-伊红(Hematoxylin-eosin,HE)染色观察肾和睾丸组织形态学改变。结果:糖皮质激素、羟基脲和腺嘌呤3种致肾阳虚药物均可使动物表现出肾阳虚症状及体征、体重下降并具有一定的致死率。与空白组相比,糖皮质激素对肾脏和睾丸组织形态影响较小,羟基脲和腺嘌呤对肾脏和睾丸等形态结构影响较大。结论:对比3个肾阳虚复合模型,羟基脲复合组模型比激素复合组和腺嘌呤复合组模型更稳定,且死亡率较低,较为理想。不过变应性鼻炎病证结合动物模型的指标评价标准尚待进一步研究。
OBJECTIVE: To establish an allergic rhinitis and kidney yang deficiency model induced by 3 different yang deficiency inducers induced by ovalbumin, allergic rhinitis and ovalbumin in guinea pig model, Ideal and stable Allergic Rhinitis (AR) disease combined with animal models. Methods: Allergic rhinitis guinea pig model was induced by Ovalbumin (OVA). At the same time, glucocorticoid, hydroxyurea or adenine were given to establish the compound model of Shen-Yang deficiency. The macro signs and deaths, the symptoms of local allergy, The contents of s Ig E and histamine in serum were detected by Enzyme-linked Immunosorbent Assay (ELISA), and morphological changes of kidney and testis were observed by hematoxylin-eosin (HE) staining. Results: All the three drugs that cause kidney - yang deficiency, such as glucocorticoid, hydroxyurea and adenine, showed the symptoms and signs of kidney - yang deficiency and the body weight of the animals decreased with a certain lethality. Compared with the blank group, glucocorticoids had little effect on the morphology of the kidney and testis, and the effects of hydroxyurea and adenine on the morphology of the kidney and testis were significant. CONCLUSIONS: Contrasting the three models of kidney-yang deficiency, the model of hydroxyurea complex is more stable than the hormone combination group and the adenine combination group, and the mortality is lower, which is more ideal. However, the combination of allergic rhinitis disease animal model index evaluation criteria remains to be further studied.
