目的:探讨内脂素(Visfatin)、IL-6与妊娠期糖尿病发生、发展的关系及相互作用。方法:选取2011年10月~2012年10月在河北省人民医院妇产科行择期剖宫产终止妊娠的患者57例,其中GDM组30例、正常妊娠组27例。ELISA法测定母血、脐血内脂素及IL-6的水平。FQ RT-PCR测定胎盘组织内脂素、IL-6 mRNA的表达水平。结果:母血及脐血Visfatin水平明显低于正常对照组(P=0.04;P=0.00);GDM组母血IL-6水平明显高于正常妊娠组(P=0.04),脐血IL-6水平高于正常妊娠组,但无统计学意义;相关性分析表明,在GDM组,母血内脂素水平与IL-6水平呈负相关(r=-0.59,P=0.01),胎盘组织中IL-6与内脂素mRNA均呈负相关(r=-0.59,P=0.045)。结论:内脂素、IL-6参与了妊娠期糖尿病的发生,且IL-6对内脂素的分泌具有一定的调节作用。
Objective: To investigate the relationship and interaction between visfatin, IL-6 and occurrence and development of gestational diabetes mellitus. Methods: From October 2011 to October 2012, 57 patients with elective cesarean termination of pregnancy in the Department of Obstetrics and Gynecology of People’s Hospital of Hebei Province, including 30 cases of GDM group and 27 cases of normal pregnancy group. ELISA method for the determination of maternal blood, umbilical blood visfatin and IL-6 levels. FQ RT-PCR determination of placental tissue adiponectin, IL-6 mRNA expression levels. Results: The levels of serum Visfatin in maternal blood and umbilical cord blood were significantly lower than those in normal controls (P = 0.04; P = 0.00). The levels of IL-6 in maternal blood of GDM patients were significantly higher than those in normal pregnant women (P = 0.04) The level of serum adiponectin was negatively correlated with IL-6 level in the GDM group (r = -0.59, P = 0.01), while in the placenta tissue, there was no significant difference IL-6 and visfatin mRNA were negatively correlated (r = -0.59, P = 0.045). Conclusion: The contents of visfatin and IL-6 are involved in the development of gestational diabetes mellitus, and IL-6 may play a regulatory role in visfatin secretion.
