目的建立同时测定蜂王浆中5-羟甲基-2-糠醛(5-HMF)、5-甲基糠醛(5-MF)、2-糠醛(F)、2-糠酸(2-OIC)、3-糠酸(3-OIC)、5-羟甲基-2-糠酸(5-HMFacid)等6种糠醛类物质含量的高效液相色谱分析方法。方法样品经去离子水提取,亚铁氰化钾和乙酸锌溶液沉淀蛋白后,用C18色谱柱分离,0.5%甲酸水溶液和甲醇梯度洗脱,在各化合物的最佳吸收波长处进行检测。结果在测试范围内,6种糠醛类物质的浓度与相应的峰面积呈线性关系,相关系数(R2)不小于0.9993。方法定量和定性检出限分别在0.07~0.26μg/m L和0.02~0.08μg/m L之间,加标回收率为86.2%~99.7%,相对标准偏差小于2.40%。结论 该方法操作简单,灵敏度高,适合蜂王浆中6种糠醛类化合物的测定。
OBJECTIVE To establish a method for the simultaneous determination of 5-HMF, 2-furfural (2-OIC), 5-methylfurfural (5-MF) - furoic acid (3-OIC), 5-hydroxymethyl-2-furoic acid (5-HMFacid) and other six kinds of furfural content of high performance liquid chromatography method. Methods The samples were extracted with deionized water. The proteins were precipitated by potassium ferrocyanide and zinc acetate solution. The proteins were separated on a C18 column and eluted with 0.5% formic acid in water and methanol. The optimum absorption wavelength of each compound was determined. Results Within the test range, the concentration of 6 furfurals showed a linear relationship with the corresponding peak area, and the correlation coefficient (R2) was not less than 0.9993. The detection limits of quantification and detection were 0.07 ~ 0.26μg / m L and 0.02 ~ 0.08μg / m L respectively. The recoveries were 86.2% ~ 99.7% with relative standard deviations less than 2.40%. Conclusion The method is simple, sensitive and suitable for the determination of six furfural compounds in royal jelly.
