目的:建立高效液相色谱—蒸发光散射检测器(HPLC—ELSD)测定冠脉通片中黄芪甲苷含量的方法。方法:采用ELSD以黄芪甲苷为对照品对冠脉通片中的黄芪甲苷进行HPLC分析,色谱柱:ZORBAXSB—C18;流动相:乙腈—水(32:68):柱温:30℃;流速:1.0ml/min:ELSD检测器漂移管温度:105℃,气体(空气)流速:2.7ml/min;结果冠脉通片中黄芪甲苷含量测定线性范围为1.0432~7.3024ug(r=0.9994),平均加样回收率为97.5%,RSD= 1.20%。结论本测定方法简便可行、重复性好,可用于冠脉通片的质量控制。
Objective: To establish a high performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD) method for the determination of astragaloside in coronary artery patency. METHODS: HPLC analysis of astragaloside IV in coronary artery bypass tablets was performed with ELSD using astragaloside as the reference substance. Column: ZORBAXSB-C18; mobile phase: acetonitrile-water (32:68): column temperature: 30 °C; Flow rate: 1.0ml/min: ELSD detector drift tube temperature: 105°C, gas (air) flow rate: 2.7ml/min; Results The linear range of astragaloside IV in coronary artery patency was 1.0432~7.3024ug (r=0.9994) The average sample recovery was 97.5% with RSD = 1.20%. Conclusion The method is simple, feasible, and reproducible. It can be used for the quality control of coronary circulation.
