目的 :研究三七总甙对NB4细胞促凝活性和组织因子表达及诱导分化作用的影响。方法 :用三七总甙处理NB4细胞 ,测定复钙时间观察NB4细胞的促凝活性 (procoagulantactivity ,PCA) ,RT PCR检测TF mRNA的表达 ;采用四唑蓝 (MTT)法检测三七总甙对NB4细胞增殖抑制作用 ;以NBT还原实验、细胞形态学观察及流式细胞术分析研究三七总甙对NB4细胞诱导分化的影响。结果 :(1)不同浓度三七总甙均可显著延长NB4细胞的复钙时间 ,降低NB4细胞的PCA水平 ,呈时间浓度依赖性 ,同时下调组织因子TF mRNA的表达 ;(2 )三七总甙可部分抑制NB4细胞的增殖 ;(3)三七总甙可提高NB4细胞NBT还原能力(P <0 0 5 ) ,细胞形态方面显示单核 /巨噬细胞分化趋势。结论 :三七总甙可降低NB4细胞的促凝活性及TF mRNA的表达 ,诱导NB4细胞部分分化 ,有望作为新的抗凝药物。
Objective: To study the effects of Panax Notoginseng saponins on procoagulant activity, tissue factor expression and differentiation in NB4 cells. Methods: NB4 cells were treated with Panax notoginsenoside, the procoagulant activity (PCA) of NB4 cells was determined by calcium complexation assay, and the expression of TF mRNA was detected by RT-PCR. The content of Panax notoginseng was detected by MTT assay NB4 cells proliferation inhibition; NBT reduction experiments, morphological observation and flow cytometry analysis of Panax Notoginseng NB4 cells induced differentiation. Results: (1) Panax notoginseng saponins at different concentrations significantly prolonged the time of calcium-resuscitation in NB4 cells and decreased the PCA level in NB4 cells in a time-dependent manner, while down-regulated the expression of tissue factor TF mRNA. (2) Glycosides can partially inhibit the proliferation of NB4 cells; (3) Panax notoginseng glycosides can improve NB4 cell NBT reduction ability (P <0 05), cell morphology showed monocyte / macrophage differentiation trend. Conclusion: Panax notoginseng saponins can reduce the procoagulant activity of NB4 cells and the expression of TF mRNA, and induce the partial differentiation of NB4 cells, which is expected to be a new anticoagulant drug.
