目的研究刺激新生大鼠心肌细胞(NRCMs)整联蛋白及dysferlin是否增加NRCMs对某些大分子物质的摄取及其机制。方法以甘氨酸-精氨酸-甘氨酸-天冬酰胺-色氨酸序列(Gly-Arg-Gly-Asp-Ser,RGD)刺激NRCMs整联蛋白及dysferlin,同时加入不同浓度德克萨斯红标记的卵蛋白(OTR)、右旋糖苷(DTR),孵育8、16、24 h,研究NRCMs对OTR、DTR的摄取及Dysferlin面积。n=30。结果 RGD增加NRCMs对OTR、DTR的摄取(与对照组比较P<0.001)。并与孵育时间呈正相关(P<0.001)。OTR、DTR的摄取与其浓度呈正相关(P<0.001)。RGD组Dysferlin面积为无RGD组的3倍。结论 RGD刺激整联蛋白及dysferlin增加NRCMs对OTR、DTR的摄取。Dysferlin参与RGD诱导的心肌细胞膜损伤的修复过程。
Objective To investigate whether the stimulation of integrin and dysferlin in neonatal rat cardiomyocytes (NRCMs) increases the uptake of some macromolecules by NRCMs and its mechanism. Methods NRCMs integrins and dysferlin were stimulated with glycine-arginine-glycine-asparagine-tryptophan (RGD), and different concentrations of Texas Red labeled (OTR) and dextran (DTR) for 8, 16, and 24 hours. The uptake of OTR and DTR by NRCMs and the area of Dysferlin were studied. n = 30. Results RGD increased the uptake of OTR and DTR by NRCMs (P <0.001 compared with the control group). And was positively correlated with incubation time (P <0.001). OTR, DTR intake and its concentration was positively correlated (P <0.001). The Dysferlin area in RGD group was 3 times of that in RGD-free group. Conclusion RGD stimulates integrin and dysferlin to increase the uptake of OTR and DTR by NRCMs. Dysferlin is involved in RGD-induced repair of myocardial cell membrane damage.
