目的分析比较代谢相关蛋白在成人和胎儿肝脏中的分布及其丰度。方法使用含19个线粒体单抗的蛋白芯片分析4种肝组织总蛋白样本(成人肝组织匀浆蛋白,胎肝匀浆蛋白,成人肝线粒体蛋白和胎肝线粒体蛋白),通过生物信息学工具对蛋白表达丰度进行比较,探讨蛋白质代谢相关的途径。结果醛氧化酶和羰基还原酶在成人肝线粒体中的表达较胎儿分别上调2.6和1.7倍。皮质类固醇11-β-脱氢酶同工酶1,环氧化物水解酶1和纤维蛋白原β链蛋白则分别下调1.7,1.9和2.2倍。环氧水解酶1和谷胱甘肽转移ω-1在成人和胎儿肝匀浆样本中存在显著差异。结论肝脏代谢相关蛋白的表达在成人肝脏和胚胎肝脏中存在显著的差别。该研究将有助于更好地理解代谢蛋白的发生和发展,并确定肝脏代谢标志物。
Objective To analyze and compare the distribution and abundance of metabolic related proteins in adult and fetal liver. Methods Total protein samples of four kinds of liver tissues (adult liver homogenate protein, fetal liver homogenate protein, adult liver mitochondrial protein and fetal liver mitochondrial protein) were analyzed using a protein chip containing 19 mitochondrial MAbs. Bioinformatics tools Protein abundance were compared to explore protein metabolism related pathways. Results The expression of aldehyde oxidase and carbonyl reductase in adult liver mitochondria were up-regulated by 2.6 and 1.7 times respectively compared with that of fetus. The corticosteroids 11-β-desaturase isoenzyme 1, epoxide hydrolase 1 and fibrinogen β-chain protein were down-regulated 1.7, 1.9 and 2.2 fold, respectively. There is a significant difference in the samples of adult and fetal liver homogenates between the hydroperoxidase 1 and the glutathione-transferred omega-1. Conclusion There is a significant difference in the expression of hepatic metabolism-related proteins between adult liver and embryonic liver. The study will help to better understand the occurrence and development of metabolic proteins and identify liver metabolic markers.
