目的 建立简便、灵敏的GC MS定量分析方法 ,以测定人血浆中的盐酸丙哌维林 (P4)及其主要代谢产物丙哌维林氮氧化物 (P4NO)。方法 血浆经碱化 (pH9) ,氯仿提取。以地西泮为内标 ,GC MS选择离子检测。结果 盐酸丙哌维林浓度在 6 .2 5~ 40 0ng·ml- 1 内具有良好的线性关系 ,最低检出浓度约为 3.13ng·ml- 1 。血浆中丙哌维林的回收率为 81.7%~ 87.5 % ,日内RSD <7.7% ,日间RSD <10 .4%。结论 本法适用于盐酸丙哌维林的生物利用度研究和血药浓度监测。
Objective To establish a simple and sensitive GC MS quantitative method for the determination of propiverine hydrochloride (P4) and its metabolite propiverine nitrogen oxide (P4NO) in human plasma. Methods Plasma alkalinization (pH9), chloroform extraction. With diazepam as internal standard, GC MS selected ion detection. Results The concentration of propiverine hydrochloride had a good linear relationship in the range of 6.52 ~ 40.0ng · ml-1 with the minimum detectable concentration of 3.13ng · ml-1. The plasma recovery of propiverine was between 81.7% and 87.5% with RSD <7.7% and daytime RSD <10.4%. Conclusion This method is suitable for bioavailability and plasma concentration monitoring of propiverine hydrochloride.
