目的探讨模拟失重30 d后再超重对猴舌黏膜组织中IL-17A及IL-23 m RNA表达的影响。方法 23只雄性猕猴采用随机数字表法分为4组,对照组(A组,n=3)、失重组(B组,n=3)、超重组(C组,n=3)、失重30 d后再超重组(D组),D组根据再超重载荷量又分为4个亚组。实验期间,各组喂养条件相同,D4组在实验结束动物恢复9 d后进行解剖取材,其余各组均在实验结束动物恢复1 d后进行解剖取材。通过Q-PCR技术检测猕猴舌黏膜组织中IL-17A和IL-23 m RNA的表达。结果超重组、失重后再超重组13 Gx组和15 Gx组中,IL-17A及IL-23 m RNA的表达显著高于对照组和失重组(P<0.05);IL-17A m RNA在失重后再超重13 Gx组和15 Gx组中的表达无明显差异;与失重后再超重13 Gx组相比,恢复9 d组的IL-17A及IL-23 m RNA表达下降,IL-17A差异有统计学意义(P<0.05),但IL-23 m RNA不具有统计学意义。对猴舌黏膜组织中的IL-17A与IL-23m R NA进行了Spea r ma n秩相关性分析,I L-17A m R NA表达与IL-23 m RNA表达之间具有高度的相关性(r=0.923,P<0.0001)。结论在模拟失重30 d后再超重作用下,猴舌黏膜组织中IL-17A和IL-23表达明显增强。
Objective To investigate the effects of overweight on IL-17A and IL-23 mRNA expression in monkey tongue mucosa 30 days after simulated weightlessness. Methods Twenty-three male macaques were randomly divided into 4 groups: control group (group A, n = 3), weightless group (group B, n = 3), overweight group (group C, n = 3) d and then overweight group (D group), D group is divided into four subgroups according to the overweight load. During the experiment, the feeding conditions were the same in each group. The animals in the D4 group recovered 9 days after the end of the experiment, and the other groups were anatomized at the end of the experiment. The expression of IL-17A and IL-23m RNA in the mucosa of kiwi was detected by Q-PCR. Results The expressions of IL-17A and IL-23 mRNA in overweight group, weight-loss and then over-weight group were significantly higher than those in control group and weight-loss group in 13 Gx group and 15 Gx group (P <0.05) There was no significant difference in the expression of IL-17A and IL-23mRNA between the 13-Gx group and the 15-Gx group after weightless overweight Statistical significance (P <0.05), but IL-23 m RNA was not statistically significant. Spearman rank correlation analysis of IL-17A and IL-23m RNA in monkey tongue mucosa showed a high correlation between IL-17A mRNA and IL-23 mRNA expression ( r = 0.923, P <0.0001). Conclusion The expressions of IL-17A and IL-23 in the mucosa of monkey tongue were significantly increased after 30 days of simulated weightlessness.
