目的通过分析不明原因复发性流产(URSA)患者,淋巴细胞免疫治疗(LIT)后外周血及子宫内膜CD3~+CD(56+16)~+NK T比例的变化,揭示LIT作用机制。方法选取2016年4-9月该院收治的20例URSA患者,应用流式细胞仪对其LIT前后外周血及子宫内膜CD3~+CD(56+16)~+NK T细胞比例变化进行分析。结果 LIT后子宫内膜CD3~+CD(56+16)~+NK T细胞比例显著性升高(P<0.05);外周血CD3~+CD(56+16)~+NK T细胞比例LIT后未发生显著性变化(P>0.05);外周血与子宫内膜CD3+CD56+NK T细胞比例不存在相关性。结论 LIT后NK T细胞比例改变主要发生在子宫内膜局部;LIT通过改变子宫内膜局部细胞因子平衡,引起子宫局部NK T细胞比例升高,可能是LIT作用机制之一;子宫内膜NK T细胞比例是评价LIT疗效的潜在指标。
Objective To investigate the mechanism of LIT by analyzing the ratio of CD3 ~ + CD (56 + 16) ~ + NK T in peripheral blood and endometrium of patients with recurrent spontaneous abortion (URSA) and lymphocyte immunotherapy (LIT) Methods Twenty patients with URSA admitted to our hospital from April to September in 2016 were enrolled in this study. Flow cytometry was used to analyze the ratio of CD3 ~ + CD (56 + 16) ~ + NK T cells in peripheral blood and endometrium before and after LIT . Results The proportion of CD3 ~ + CD (56 + 16) ~ + NK T cells in endometrium was significantly increased after LIT (P <0.05), and the ratio of CD3 ~ + CD (56 + There was no significant change (P> 0.05). There was no correlation between the proportion of CD3 + CD56 + NK T cells in peripheral blood and endometrium. Conclusions The proportion of NK T cells after LIT mainly changes in the part of endometrium. LIT can increase the percentage of NK T cells in the uterus by changing the balance of cytokines in the endometrium, which may be one of the mechanisms of LIT. Endometrium NK T Cell ratio is a potential measure of efficacy of LIT.
