目的 :建立多天线癌胚抗原 (multipleantennaecarcinoembryonicantigen ,MA CEA)的快速定量检测法 ,并研究肿瘤患者血清中多天线癌胚抗原的水平。方法 :采用神经氨酸酶 (neuramin-idase ,NMD)降解MA CEA糖链外侧的唾液酸。采用蔓陀萝凝集素 (daturastramoniumagglutinin ,DSA)和CEA抗体 ,建立检测MA CEA的链霉亲和素 生物素系统酶免疫分析法 (ABC EIA) ,分别检测 2 39例肺癌、胃癌、肝癌、结肠癌和卵巢癌患者血清中MA CEA的水平 ,同时采用ELISA检测血清CEA的水平。结果 :血清MA CEA的水平在CEA阳性的肺癌患者中明显升高 ,75例肺癌患者血清MA CEA的阳性率为 14 .7% ,而CEA水平升高的肺癌阳性率为 4 7.3% ;但胃癌、肝癌、结肠癌和卵巢癌患者 ,无论CEA水平是否升高 ,均未检测到MA CEA水平的升高。结论 :建立了一种可定量检测肿瘤患者血清MA CEA的方法 ,为肺癌的鉴别诊断及其他多天线糖链肿瘤标志物的研究奠定了基础
OBJECTIVE: To establish a rapid quantitative assay for multiple antenna carcinoembryonic antigen (MA CEA) and study the level of multi-antenna carcinoembryonic antigen in the serum of tumor patients. Methods: The neuraminidase (neuramin-idase, NMD) was used to degrade the sialic acid outside the MA CEA sugar chain. The streptavidin-biotin enzyme immunoassay (ABC EIA) was established to detect MA CEA by using daturastramoniumagglutinin (DSA) and CEA antibody, and 239 cases of lung cancer, gastric cancer, liver cancer, colon cancer And the level of MA CEA in the serum of patients with ovarian cancer, meanwhile, the level of serum CEA was detected by ELISA. Results: Serum MA CEA level was significantly higher in patients with CEA positive lung cancer. The positive rate of MA CEA in 75 patients with lung cancer was 14.7%, while the positive rate of CEA was 43.3% , Liver cancer, colon cancer, and ovarian cancer, no increase in MA CEA was detected regardless of whether CEA levels were elevated or not. Conclusion: A method for quantitative determination of serum MA CEA in patients with cancer was established, which laid the foundation for the differential diagnosis of lung cancer and other multi-antenna carbohydrate tumor markers
