目的探讨丹参酮ⅡA(TanshinoneⅡA,TanⅡA)对小鼠骨髓瘤细胞SP2/0的生长抑制与诱导凋亡作用。方法选用小鼠骨髓瘤细胞系SP2/0,运用光镜、MTT法、DNA琼脂糖凝胶电泳分析SP2/0细胞增殖及凋亡的改变。结果经丹参酮ⅡA处理后,SP2/0细胞增殖明显被抑制,当4.0μg/mL TanⅡA处理SP2/0细胞72 h,增殖抑制率达(79.4±2.2)%,镜下可见凋亡细胞的形态学改变,细胞DNA片断化(呈现“梯状”断裂现象)。结论TanⅡA能有效地抑制小鼠骨髓瘤细胞系SP2/0增殖,诱导SP2/0细胞凋亡。
Objective To investigate the effect of TanshinoneⅡA (TanⅡA) on the growth inhibition and apoptosis induction of mouse myeloma cells SP2 / 0. Methods The mouse myeloma cell line SP2 / 0 was selected. The proliferation and apoptosis of SP2 / 0 cells were analyzed by light microscopy, MTT and DNA agarose gel electrophoresis. Results After treated with tanshinone ⅡA, the proliferation of SP2 / 0 cells was significantly inhibited. When 4.0 μg / mL TanⅡA treated SP2 / 0 cells for 72 h, the proliferation inhibition rate was (79.4 ± 2.2)%. The morphology of apoptotic cells Change, cell DNA fragmentation (showing “ladder ” fracture phenomenon). Conclusion TanⅡA can effectively inhibit the proliferation of mouse myeloma cell line SP2 / 0 and induce the apoptosis of SP2 / 0 cells.
