目的:建立超高效液相色谱法测定榄香烯脂质体的药物含量及包封率。方法:色谱柱为XR-ODSⅡ柱(2.2μm,75mm×3.0mm),流动相为乙腈-水(80∶20),柱温为40℃,检测波长为210nm,流速为1.0mL·min-1。结果:色谱条件下榄香烯与辅料、溶剂峰分离良好,β-榄香烯在1~100μg·mL-1范围内线性关系良好。榄香烯脂质体平均包封率为(99.8±0.4)%。结论:超高效液相色谱法可用于检测榄香烯脂质体的含量和包封率,简单、快速且准确。
OBJECTIVE: To establish a method for the determination of elemene liposomes in drug content and entrapment efficiency by ultra performance liquid chromatography. Methods: The column was XR-ODSⅡ (2.2μm, 75mm × 3.0mm). The mobile phase was acetonitrile-water (80:20). The column temperature was 40 ℃, the detection wavelength was 210nm and the flow rate was 1.0mL · min -1 . Results: Under the chromatographic conditions, the separation of elemene from the excipients and solvent peaks was good. The linear relationship of β-elemene in the range of 1 ~ 100μg · mL-1 was good. The average entrapment efficiency of elemene liposomes was (99.8 ± 0.4)%. Conclusion: Ultra Performance Liquid Chromatography can be used to detect the content and entrapment efficiency of elemene liposomes, which is simple, rapid and accurate.
