以质粒pIJ702和pIJ699为载体,构建了两个带有透明颤菌(Vitreoscila)血红蛋白(VHb)基因(Vgb)的大肠杆菌-链霉菌穿梭质粒并转化E.coliHB101、变青链霉菌(S.lividans)TK64及蒽环类抗生素阿柔比星(阿克拉菌素)产生菌——加利利链霉菌(S.galilaeus)ATCC31133,经CO结合差光谱法、SDS-PAGE蛋白电泳分析表明Vgb在大肠杆菌和链霉菌中得到表达。通过改变培养条件研究溶氧对Vgb表达的影响,结果表明在低溶氧的情况下,VHb表达量增加。同时发现,在低氧浓度时,VHb的表达有利于链霉菌菌体的生长,其菌体生长量高于同样条件下的对照菌株(Vgb-)17%~29%。
Two E. coli-Streptomyces shuttle plasmids carrying the Vitreoscilla hemoglobin (VHb) gene (Vgb) were constructed using plasmids pIJ702 and pIJ699 as vectors and transformed into E. coli. coli HB101, S.lividans TK64 and anthracycline-producing strain acylase (S. alannamei) producing strain S. galilaeus ATCC31133, SDS-PAGE protein electrophoresis analysis showed that Vgb was expressed in E. coli and Streptomyces. The effects of dissolved oxygen on the expression of Vgb were studied by changing the culture conditions. The results showed that the expression of VHb was increased in the presence of low dissolved oxygen. At the same time, it was found that the expression of VHb was conducive to the growth of Streptomyces sp. At low oxygen concentration. The growth of the strain was higher than that of the control strain (Vgb-) under the same conditions by 17% -29%.
