目的建立荷人胃肠间质瘤裸鼠动物模型,并对其生物学行为进行初步探讨。方法BALB/C/nu/nu品系裸鼠皮下接种人胃肠间质瘤组织块,观察成瘤情况及移植瘤病理组织学特点,并用免疫组织化学SP方法测定移植瘤的CD117的表达情况。结果移植瘤生长稳定,30只裸鼠荷瘤成功26只,荷瘤成功率86%,病理组织学与免疫组织化学均证实移植瘤保持了人胃肠间质瘤的生物学特征。结论采用BALB/C/nu/nu品系裸鼠皮下接种人胃肠间质瘤组织块的方法初步建立了人胃肠间质瘤的裸鼠皮下移植瘤模型,为进一步研究胃肠间质瘤的侵袭生长特性奠定了基础。
Objective To establish an animal model of human gastrointestinal stromal tumor in nude mice and to investigate its biological behavior. Methods BALB / C / nu / nu nude mice were inoculated subcutaneously with human gastrointestinal stromal tumor tissue to observe tumor formation and histopathological features of the transplanted tumor. The expression of CD117 in the transplanted tumor was detected by immunohistochemical SP method. Results The growth of xenografts was stable. Twenty-six nude mice were successful in tumor-bearing and the success rate of tumor-bearing was 86%. Both histopathology and immunohistochemistry confirmed that the xenografts maintained the biological characteristics of human gastrointestinal stromal tumors. Conclusions A subcutaneous xenograft tumor model of human gastrointestinal stromal tumor was established by subcutaneous inoculation of human gastrointestinal stromal tissue in BALB / c / nu / nu nude mice. Invasion of growth characteristics laid the foundation.
