目的:建立高效液相色谱法(HPLC)测定葱白提取物软胶囊中β-谷甾醇的含量。方法:采用Cosmosil C18色谱柱(4.6mm×250mm,5μm),以甲醇-水(98∶2)为流动相,流速1.0mL·min-1,紫外检测波长210nm;柱温30℃。结果:β-谷甾醇在20.2~60.6mg·L-1范围内峰面积与浓度呈良好的线性关系,回归方程为Y=0.0659X+0.3657,r=0.9993(n=6)。平均回收率为99.77%(n=9,RSD为1.57%)。结论:高效液相色谱法能够精确的检测葱白提取物软胶囊中β-谷甾醇的含量,应用该方法精密度高,重复性好。
Objective: To establish a high performance liquid chromatography (HPLC) determination of β-sitosterol in soft white extract of Ephedra. Methods: Cosmosil C18 column (4.6 mm × 250 mm, 5 μm) was used with mobile phase of methanol-water (98: 2) as mobile phase at a flow rate of 1.0 mL · min-1. UV detection wavelength was 210 nm. Results: The peak area of β-sitosterol in the range of 20.2-60.6 mg · L-1 showed a good linear relationship with the concentration. The regression equation was Y = 0.0659X + 0.3657, r = 0.9993 (n = 6). The average recovery was 99.77% (n = 9, RSD 1.57%). Conclusion: HPLC method can accurately detect the content of β-sitosterol in the soft capsule of scallion extract. The precision and repeatability of this method are good.
