目的 研究紫杉醇能否诱导ACC 2细胞凋亡 ,以及凋亡诱导与G2 /M期的关系。方法紫杉醇不同浓度、不同作用时间分别处理ACC 2细胞 ,通过流式细胞仪、荧光显微镜、透射电镜和DNA凝胶电泳 ,研究紫杉醇诱导ACC 2细胞的凋亡及其与G2 /M期阻滞的关系。结果 荧光染色观察到凋亡细胞形态 ,电镜观察到不同时期凋亡细胞超微结构的典型形态。流式细胞仪DNA含量分析 :紫杉醇在 5 0nmol/L作用 48h、72h ,亚G1峰分别是 17 13%、16 2 6 %。延长药物作用时间、增加药物浓度均会增强细胞G2 /M期阻滞 ,但只有出现G2 /M期明显阻滞后 ,经过一定时间才出现凋亡。结论紫杉醇能够诱导ACC 2细胞产生凋亡 ;G2 /M期阻滞能诱导ACC 2细胞凋亡。
Objective To investigate whether paclitaxel can induce apoptosis of ACC 2 cells and the relationship between apoptosis induction and G2 / M phase. Methods Paclitaxel (ACC 2) cells were treated with different concentrations of paclitaxel at different concentrations for different time periods. The apoptosis of ACC 2 cells induced by paclitaxel and its relationship with G2 / M arrest were studied by flow cytometry, fluorescence microscopy, transmission electron microscopy and DNA gel electrophoresis. relationship. Results The morphology of apoptotic cells was observed by fluorescence staining. The ultrastructural changes of apoptotic cells were observed under electron microscope. Flow cytometry DNA content analysis: paclitaxel in the role of 50nmol / L 48h, 72h, sub G1 peak were 17 13%, 16 2 6%. Prolonged drug action time and increased drug concentration increased cell G2 / M arrest, but apoptosis occurred only after a certain period of G2 / M arrest. Conclusion Paclitaxel can induce apoptosis in ACC 2 cells and G2 / M arrest can induce apoptosis of ACC 2 cells.
