目的:观察青藤碱对NZB/WF1代狼疮鼠外周血CD4+T细胞及Bax、Bcl-2mRNA表达的影响。方法:随机选择NZB/WF1代狼疮鼠32只分为4组:对照组、青藤碱低、中、高剂量组。分别采用流式细胞计数法和realtimeRT-PCR法检测NZB/WF1代狼疮鼠外周血中CD4+T细胞数量及Bax、BCL-2mRNA表达。结果:青藤碱干预三周后,NZB/WF1代狼疮鼠外周血CD4+T细胞数量升高;BaxmRNA表达明显升高,Bcl-2mRNA表达明显降低。结论:青藤碱能减轻NZB/WF1代狼疮鼠的T细胞等病变。
OBJECTIVE: To observe the effects of sinomenine on the expression of CD4+ T cells and Bax and Bcl-2 mRNA in NZB/WF1 lupus erythematosus mice. METHODS: Thirty-two NZB/WF1 lupus mice were randomly divided into 4 groups: control group, low, middle and high doses of sinomenine. Flow cytometry and realtime RT-PCR were used to detect the number of CD4+ T cells and the expression of Bax and BCL-2 mRNA in NZB/WF1 lupus mice. RESULTS: After three weeks of intervention with sinomenine, the number of CD4+ T cells in the peripheral blood of NZB/WF1 lupus mice increased; the expression of Bax mRNA was significantly increased and the expression of Bcl-2 mRNA was significantly decreased. Conclusion: Sinomenine can reduce T cell and other diseases in NZB/WF1 lupus mice.
