目的探讨白细胞介素18(IL-18)基因编码区第105位点基因多态性与草酸钙肾结石病的关系。方法选取2010年6月至2011年10月在深圳市第二人民医院泌尿外科就诊的60例尿路结石患者列为草酸钙结石组,将同期来医院健康查体的志愿者60例作为对照组,用聚合酶链反应-限制性片段长度多态性方法检测所有研究对象的IL-18基因编码区第105位点基因型。结果经过TaqⅠ酶切的IL-18 DNA片段可分为三种基因型:AA(148 bp)、AC(148、123、25 bp)及CC(123、25 bp),草酸钙结石组AC+CC基因型(46.6%,28/60)与对照组(21.6%,13/60)相比有统计学意义(P<0.01),等位基因C在草酸钙结石组(25%,30/120)和对照组(10.8%,13/120)频率分布相比有统计学意义(P<0.01)。结论 IL-18基因编码区第105位点基因型多态性与草酸钙肾结石病有相关性,IL-18基因编码区第105位点C等位基因可作为肾石病患者草酸钙结石危险因素的标志。
Objective To investigate the relationship between gene polymorphism at position 105 of interleukin 18 (IL-18) gene and calcium oxalate nephrolithiasis. Methods Sixty patients with urolithiasis admitted to Department of Urology, Second People’s Hospital of Shenzhen from June 2010 to October 2011 were classified as oxalic acid calcium stones group. Sixty volunteers who were hospitalized during the same period were selected as control group The genotype of site 105 of coding region of IL-18 gene was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results There were three genotypes of IL-18 DNA fragment digested with TaqⅠ: AA (148 bp), AC (148,123,25 bp) and CC (123,25 bp) The allele C was significantly higher in the calcium oxalate stones group (25%, 30/120) than in the control group (21.6%, 13/60) (P <0.01) And the control group (10.8%, 13/120) frequency distribution was statistically significant (P <0.01). Conclusion The polymorphism of IL-18 gene coding site 105 is associated with calcium oxalate nephrolithiasis. The C allele of IL-18 gene coding region may be used as the risk of calcium oxalate stones in patients with nephrolithiasis The sign of the factor.
