目的:探讨三氧化二砷(As2O3)诱导人胆囊癌(GBC)细胞凋亡及对干细胞标志物CD133表达的作用。方法:用Hoechest染色、流式细胞仪和DNA凝胶电泳研究As2O3诱导GBC细胞凋亡及对CD133+GBC细胞的作用。Western印迹法检测CD133蛋白表达,RT-PCR检测CD133 mRNA表达。结果:As2O3能诱导GBC细胞凋亡、减少CD133+GBC细胞的数量,同时能降低GBC细胞CD133的蛋白及mRNA表达。结论:As2O3可能通过抑制干细胞标志物CD133表达而实现其对体外人GBC细胞的诱导凋亡作用。
Objective: To investigate the effect of arsenic trioxide (As2O3) on the apoptosis of human gallbladder carcinoma (GBC) cells and its effect on the expression of stem cell marker CD133. Methods: Hoechst staining, flow cytometry and DNA gel electrophoresis were used to study the apoptosis of GBC cells induced by As2O3 and its effect on CD133 + GBC cells. CD133 protein expression was detected by Western blotting and CD133 mRNA expression was detected by RT-PCR. Results: As2O3 could induce apoptosis of GBC cells, decrease the number of CD133 + GBC cells, and decrease the protein and mRNA expression of CD133 in GBC cells. Conclusion: As2O3 may induce apoptosis of human GBC cells in vitro by inhibiting the expression of stem cell marker CD133.
