目的:观察背部循经推拿对亚健康模型大鼠海马脑源性神经营养因子(BDNF)基因和蛋白表达的影响,初步探讨背部循经推拿调理亚健康状态的作用机制。方法:60只Wistar大鼠随机分为正常组、模型组和推拿干预组,后两组予束缚应激,推拿干预组予背部循经推拿。实验结束后,采用实时荧光定量PCR方法检测大鼠海马BDNF mRNA表达,免疫印迹技术检测大鼠海马BDNF蛋白表达水平。结果:与空白组比较,模型组大鼠海马BDNF mRNA及其蛋白表达水平下降,差异有显著性(P<0.05)。与模型组比较,推拿干预组大鼠海马BDNF mRNA及其蛋白表达水平均明显升高,差异有显著性(P<0.05)。结论:背部循经推拿手法能上调亚健康模型大鼠海马区BDNF蛋白表达水平,这是推拿调治亚健康状态的作用途径之一。
OBJECTIVE: To observe the effect of back massage on the gene and protein expression of brain-derived neurotrophic factor (BDNF) in hippocampus of sub-health rats, and to explore the mechanism of back-passage massage in regulating sub-health status. Methods: Sixty Wistar rats were randomly divided into normal group, model group and massage intervention group, the latter two groups were given restraint stress, and the massage intervention group was given back massage. After the experiment, BDNF mRNA expression in the hippocampus was detected by real-time fluorescence quantitative PCR. BDNF protein expression in the hippocampus was detected by Western blotting. Results: Compared with the blank group, the expression of BDNF mRNA and protein in the hippocampus of model group decreased significantly (P <0.05). Compared with model group, the expression of BDNF mRNA and protein in hippocampus of rats in massage intervention group were significantly increased, the difference was significant (P <0.05). CONCLUSION: BDNF protein expression in the hippocampus of sub-health rats can be increased by the manipulation of the back through the hands and feet, which is one of the ways to modulate the sub-health state of massage.
