目的探讨亚砷酸钠(NaAsO2)对人皮肤角质形成细胞(HaCaT)增殖、细胞周期及活性氧(ROS)生成影响。方法以0、25、50μmol/L NaAsO2作用于HaCaT细胞6 h后,以Alamar Blue还原法检测细胞增殖水平;以流式细胞仪检测细胞周期及ROS水平。结果 25、50μmol/L NaAsO2组细胞增殖水平分别为(93.5±1.18)%、(82.9±2.64)%,明显低于对照组(100±0.51)%(P<0.05);G2/M期细胞数分别为(19.15±0.29)%、(18.12±1.35)%,明显高于对照组(15.24±0.04)%(P<0.05);ROS水平分别为(128.61±6.23)%、(152.92±7.25)%,明显高于对照组(100.00±3.45)%(P<0.05)。结论 NaAsO2作用可引起HaCaT细胞ROS含量增高,同时G2/M期细胞数增多,但G2/M期细胞比例增高并不是由细胞增殖作用所引起。
Objective To investigate the effects of sodium arsenite (NaAsO2) on proliferation, cell cycle and reactive oxygen species (ROS) production of human keratinocytes (HaCaT). Methods HaCaT cells were treated with 0, 25 and 50μmol / L NaAsO2 for 6 h. The cell proliferation was detected by Alamar Blue reduction. The cell cycle and ROS levels were measured by flow cytometry. Results The proliferation of 25 and 50μmol / L NaAsO2 groups were (93.5 ± 1.18)% and (82.9 ± 2.64)%, respectively, which were significantly lower than those of the control group (100 ± 0.51)% (19.15 ± 0.29)% and (18.12 ± 1.35)%, respectively, which were significantly higher than that of the control group (15.24 ± 0.04)% (P <0.05). The ROS levels were (128.61 ± 6.23)% and (152.92 ± 7.25) , Which was significantly higher than that of the control group (100.00 ± 3.45)% (P <0.05). Conclusion NaAsO2 can induce the increase of ROS content in HaCaT cells and increase the number of cells in G2 / M phase. However, the increased proportion of cells in G2 / M phase is not caused by cell proliferation.
