目的 探讨 TNFα 引起的血管内皮细胞 (VEC)凋亡在皮肤撕脱伤早期血栓形成中的作用 .方法 分 2组 ,实验组 :内皮细胞 (EC) +TNFα,拮抗组 :用 EC+TNFα+TNFα抗体处理 .采用流式细胞仪分别测定实验组 TNFα不同时间、不同浓度以及拮抗组 EC凋亡率 .结果 EC凋亡率随 TNFα作用时间增加 ,0~ 48h EC凋亡率为 (2 .6± 0 .42 ) %~ (4 0 .5±8.3) % ;EC凋亡率也随 TNFα浓度而增加 ,0~ 30 0 0 u· m L- 1EC凋亡率分别为 (2 .6± 0 .42 ) %~ (13.7± 2 .6 ) % ;TNFα抗体明显拮抗 TNFα诱导的 EC凋亡 (P<0 .0 1) .结论 TNFα可引起 EC的凋亡和死亡 ,具有时间依赖性和浓度依赖性 ,人重组 TNFα单抗可消除因 TNFα引起的毒性作用
Objective To investigate the role of apoptosis induced by TNFα in the early thrombosis of skin avulsion in rats.Methods Two groups were divided into experimental group (EC + TNFα) and antagonist group (EC + TNFα + TNFα Antibody treatment.Flow cytometry was used to determine the apoptotic rate of TNFα in the experimental group at different time and different concentrations as well as in the antagonistic group.Results The apoptotic rate of EC increased with the time of TNFα and the apoptotic rate of EC at 0-48h was (2.6 ± 0.42% ~ (40.5 ± 8.3)%. The apoptosis rate of EC also increased with the concentration of TNFα, and the apoptotic rates of 0-30 000 μm L-1EC were (2.6 ± 0.)%. 42)% ~ (13.7 ± 2 .6)%; TNFαantibodies significantly antagonized the TNFαinduced EC apoptosis (P0.01) .Conclusion TNFα can cause apoptosis and death of EC in a time-dependent and concentration-dependent manner Sex, human recombinant TNFα monoclonal antibody can eliminate the toxic effects caused by TNFα
