rn目的探讨氯胺酮对脂多糖所致大鼠抑郁样症状的影响。方法 30只雄性Wistar大鼠随机均分为对照(C)组、生理盐水(S)组和氯胺酮(K)组。K组及S组大鼠腹腔注射1mg/kg脂多糖,24h后分别注射10mg/kg氯胺酮和等体积生理盐水。30min后,三组大鼠行强迫游泳实验6min,记录后5min不动时间。立即处死大鼠,取前额皮层采用ELISA法测定IL-1β和IL-6的含量。结果与C组相比,S组大鼠强迫游泳不动时间显著延长(P“,”Objective To investigate the effects of ketamine on lipopolysaccharide-induced depression-like symptoms in rats.Methods Thirty male Wistar rats were equally randomized into 3 groups of C(control group),S(saline group) and K(ketamine group).Lipopolysaccharide 1 mg/kg was intraperitoneally injected in the rats of groups of K and S,which were given ketamine 10 mg/kg or same volumn of saline 24 hours later,respectively.Thirty minutes later,forced swimming test(FST) was carried out in 3 groups and the immobility time during FST was recorded.Subsequently,all rats were sacrificed and prefrontal cortex was harvested for the determinations of IL-1β and IL-6 by ELISA.Results Compared with group C,the immobility time of FST was longer(P<0.01) and expressions of IL-1β and IL-6 in prefrontal cortex were higher(P<0.05) in group S.Compared with group S,the immobility time of FST was shorter and expressions of IL-1β and IL-6 in prefrontal cortex were lower in group K(P<0.05).Conclusion Ketamine is able to ameliorate lipopolysaccharide-induced depression-like symptoms in rats,which is probably related to downregulation of IL-1β and IL-6 in the prefrontal cortex.
