目的 观察一氧化氮和血管内皮素这一对拮抗因子在糖尿病肾病中的作用。方法 用逆转录半定量聚合酶链反应 (RT PCR)法测定 12只STZ模型鼠 (糖尿病组 ) 12周时及 8只对照组鼠的肾皮质内皮素的基因表达及一氧化氮含量。结果 糖尿病组鼠肾皮质血管内皮素较正常对照组明显升高 (P <0 .0 1) ,而其一氧化氮含量却低于对照组 (P <0 .0 5 )。结论 证实在糖尿病后期血管内皮素生成增加而一氧化氮合成受阻 ,加剧了糖尿病肾脏病变的进展。
Objective To observe the effects of antagonists of nitric oxide and endothelin on diabetic nephropathy. Methods The renal cortex endothelin gene expression and nitric oxide content in 12 STZ model rats (diabetic group) at 12 weeks and 8 control rats were determined by reverse transcription semi-quantitative polymerase chain reaction (RT PCR). Results The level of endothelin in renal cortex of diabetic rats was significantly higher than that of normal controls (P <0.01), while the content of nitric oxide in diabetic rats was lower than that of control rats (P <0.05). Conclusion confirmed that in the late stage of diabetes, endothelin production increased nitric oxide synthesis blocked, exacerbated the progress of diabetic nephropathy.
