应用骨髓染色体G显带技术,在450例慢性髓性白血病(CML)检出3例隐匿型Ph易位,报道如下:1 细胞遗传学分析方法 均取患者的骨髓血,细胞数按终浓度(2~3)×10~9/ml加入含20%小牛血清或20%自体血浆的McCoy’s培养液中,在37C温箱中进行24h培养,终止培养前2h按终浓度0.08~0.1μg/ml加入秋水仙素,常规低渗、固定后,进行制片、老化、G显带,方法详见报道。每例至少分析20个中期分裂相。异常核型按人类染色体国际命名体制ISCN标准定义。对每个异常克隆拍照、放大、剪排,制备3~5个核型图。2 病例及临床资料 3例皆为男性,按张之南主编的有关标准进行诊断、分期和疗效评定。 例1,42岁,1993年7月因左肋部疼痛、低热、乏力3周,在我院门诊查白细胞(WBC)80×10~9/L,肝
Three cases of occult Ph-translocation were detected in 450 cases of chronic myelogenous leukemia (CML) using bone marrow G-banding technique. The report is as follows: 1 Cytogenetic analysis of patients with bone marrow blood cells, the number of cells by the final concentration ( 2 ~ 3) × 10 ~ 9 / ml was added to McCoy’s medium containing 20% fetal bovine serum or 20% autologous plasma and incubated in a 37C incubator for 24 hours. Abnormal karyotype according to the International Human Naming System ISCN standard definition.
