水杨酸钠降低庆大霉素所致耳蜗自由基含量的实验研究

目的 采用电子顺磁共振法 (electronparamagneticresonancespectroscopy ,EPR)观察水杨酸钠对庆大霉素注射过程中耳蜗组织羟自由基含量增加的预防作用。方法 耳廓反射良好的豚鼠随机分成 3组 ,庆大霉素组、庆大霉素 +水杨酸钠组和正常对照组。取标本前和标本制备时向耳蜗组织加入自由基捕捉剂 5 ,5 -dimethyl -pyrroline - 1-oxide(DMPO) ,用EPR直接检测耳蜗组织中自由基含量。结果 正常对照组耳蜗组织可测出较弱的羟(OH)自由基信号 ,庆大霉素注射后 ,自由基信号明显增强 (P <0 .0 1) ,水杨酸钠和庆大霉素同时注射 ,自由基信号明显减弱 (P <0 .0 1)。结论 加入自由基捕捉剂DMPO ,用EPR直接检测耳蜗组织中自由基含量是一种较有效的方法 ;水杨酸钠可部分清除庆大霉素治疗过程中产生的过量自由基 Objective To observe the preventive effect of sodium salicylate on the increase of hydroxyl radical content in cochlear during gentamicin injection by electron paramagnetic resonance spectroscopy (EPR). Methods Guinea pigs with good auricle reflex were randomly divided into 3 groups: gentamicin group, gentamicin + sodium salicylate group and normal control group. The free radical scavenger 5, 5 -dimethyl-pyrroline - 1-oxide (DMPO) was added to the cochlear tissues before and during specimen preparation. The contents of free radicals in cochlear tissue were detected directly by EPR. RESULTS: Weaker OH (OH) free radical signal was detected in cochlear tissue of normal control group. After gentamicin injection, the signal of free radical was significantly enhanced (P <0.01), and sodium salicylate and gentamycin Simultaneous injection, the free radical signal was significantly weakened (P <0.01). Conclusion The addition of free-radical scavenger DMPO and direct detection of free radical content in cochlear tissue by EPR is an effective method. Sodium salicylate can partially eliminate excess free radicals generated during gentamicin treatment
