目的建立红外辅助萃取-高效液相色谱(IRAE-HPLC)法测定丹参药材中丹酚酸B含量的方法。方法 Diamonsil C18柱(150 mm×4.6 mm,5μm),柱温为25℃,以乙腈-0.1%磷酸水溶液(22:78)为流动相;检测波长286 nm,流速1.2 mL·min~(-1)。红外辅助萃取条件的溶剂是20 mL蒸馏水,提取温度90℃,提取时间8 min。结果丹酚酸B在5~100 mg·L~(-1)内线性关系关系良好(r=0.999 1),高、中、低浓度下方法的平均加样回收率为95.26%~108.47%,RSD为1.13%~2.02%(n=3),定量限(RsN=10)为0.5 mg·L~(-1),检测限(RsN=3)为0.15 mg·L~(-1)。用该方法实际测得3批丹参药材中丹酚酸B的含量可达3.9%~4.0%。结论该方法简便、快速、准确、可靠、安全、环保,适用于测定丹参药材中丹酚酸B的含量。
Objective To establish a method for the determination of salvianolic acid B in Radix Salviae Miltiorrhizae by infrared-assisted extraction-high performance liquid chromatography (IRAE-HPLC). Methods Diamonsil C18 column (150 mm × 4.6 mm, 5 μm) was used with a mobile phase of acetonitrile-0.1% phosphoric acid (22:78) at a column temperature of 25 ℃. The detection wavelength was 286 nm and the flow rate was 1.2 mL · min -1 ). Infrared assisted extraction conditions of the solvent is 20 mL of distilled water, extraction temperature 90 ℃, extraction time 8 min. Results The linear relationship of salvianolic acid B in the range of 5-100 mg · L -1 was good (r = 0.999 1). The average recovery of salvianolic acid B was 95.26% -108.47% at high, medium and low concentrations, The RSD was 1.13% ~ 2.02% (n = 3). The limit of quantification (RsN = 10) was 0.5 mg · L -1 and the limit of detection (RsN = 3) was 0.15 mg · L -1. Using this method, the content of salvianolic acid B in 3 batches of Salviae miltiorrhizae was measured to reach 3.9% -4.0%. Conclusion The method is simple, rapid, accurate, reliable, safe and environmentally friendly and is suitable for the determination of Salvianolic acid B in Salvia miltiorrhiza.
