目的探讨大黄酸对肥胖糖尿病大鼠脂肪组织抵抗素基因表达及血浆FFA的影响。进而验证大黄酸具有改善胰岛素抵抗的作用。方法小剂量链脲佐菌素加高脂饲料诱导建立肥胖糖尿病大鼠模型,成模大鼠随机分为模型组、吡格列酮组和大黄酸组。正常组与模型组分别予生理盐水10 ml·kg~(-1)·d~(-1),吡格列酮组予吡格列酮10 mg·kg~(-1)·d~(-1),大黄酸组予大黄酸100 mg·kg~(-1)·d~(-1),分别灌胃8周。测定各组附睾脂肪垫抵抗素基因的表达、血浆FFA及血脂水平。结果与模型组相比,大黄酸组与吡格列酮组脂肪组织抵抗素的表达均下降,FFA、TC、TG、LDL-C均明显降低(P均<0.05)。结论大黄酸能降低肥胖糖尿病大鼠脂肪组织抵抗素表达,降低血浆FFA。这可能是大黄酸改善胰岛素抵抗的机制之一。
Objective To investigate the effect of rhein on adiponectin gene expression and plasma FFA in obese diabetic rats. Further verify that rhein has the effect of improving insulin resistance. Methods Small doses of streptozotocin plus high-fat diet induced the establishment of obese diabetic rat model, the model rats were randomly divided into model group, pioglitazone group and rhein group. Normal group and model group were given normal saline 10 ml · kg -1 · d -1, pioglitazone group pioglitazone 10 mg · kg -1 · d -1, The rats were given 100 mg · kg -1 d · (-1) of rhein for 8 weeks. The epididymal fat pad resistance gene expression, plasma FFA and blood lipid levels were determined. Results Compared with the model group, the expression of adiponectin in rhein group and pioglitazone group decreased, and the levels of FFA, TC, TG and LDL-C decreased significantly (all P <0.05). Conclusion Rhein can reduce adiponectin expression and decrease plasma FFA in obese diabetic rats. This may be one of the mechanisms by which rhein improves insulin resistance.
