材料和方法在山口市山口大学农学部从一株约6米高,50年树龄的山茶树(Camellia japonicaL.var.spontanea)上收集种子,茶种(薮北种)则产自金谷町的国立茶业试验场。剥去种壳之后,种子表面用70%乙醇消毒,然后在7%(W/V)的次氯酸钙中浸1小时。在不附加生长调节剂的 MS 培养基上培养1周灭菌。除去种胚后,子叶切成7—10片,然后将这些子叶切片做为外植体接种。基本培养基为 MS 无机盐,附加3%蔗糖、0.8%琼脂,盐酸
MATERIALS AND METHODS Seeds were collected from Camellia japonica L.var.spontanea at a height of about 6 meters and a 50-year-old in Agronomy Department of Yamaguchi University in Yamaguchi City. Tea industry test ground. After stripping the seed coat, the seed surface was sterilized with 70% ethanol and then soaked in 7% (w / v) calcium hypochlorite for 1 hour. Sterilized on MS medium without growth regulators for 1 week. After removing embryos, the cotyledons are cut into 7-10 pieces and these cotyledons are seeded as explants. The basal medium is MS mineral salts, supplemented with 3% sucrose, 0.8% agar, hydrochloric acid
