目的:研究蓬乱蛋白(dishevelled,DVL)-1、DVL-3在正常和尿道下裂患儿包皮组织中的表达,探讨其在尿道下裂发病机制中可能发挥的作用。方法:收集拟行尿道下裂尿道修复术患儿术中切除的包皮组织(24例)和门诊包皮环切术中切除的包皮(24例),采用免疫组化和实时定量聚合酶链反应(q PCR)方法检测两组包皮中DVL-1、DVL-3 m RNA和蛋白质的表达情况。结果:尿道下裂患儿包皮组织中DVL-1、DVL-3 m RNA表达较正常水平有显著升高,差异有统计学意义(P<0.05)。DVL-1、DVL-3蛋白主要表达在包皮皮下间充质细胞层,其中DVL-1、DVL-3蛋白在尿道下裂患儿包皮组织的表达较正常对照有明显增强趋势。结论:DVL-1、DVL-3的表达异常与尿道下裂发生有关,其介导的调控异常可能是导致尿道下裂发生的主要机制之一。
Objective: To investigate the expression of dishevelled (DVL) -1 and DVL-3 in the foreskin of normal and hypospadias children and to explore their possible role in the pathogenesis of hypospadias. Methods: The resected foreskin (24 cases) and circumcision circumcision (24 cases) in children undergoing hypospadias urethroplasty were collected. Immunohistochemistry and real - time quantitative polymerase chain reaction qPCR) were used to detect the expression of DVL-1, DVL-3 mRNA and protein in the two groups of foreskin. Results: The expression of DVL-1 and DVL-3 m RNA in the foreskin of children with hypospadias was significantly higher than the normal level, the difference was statistically significant (P <0.05). The expression of DVL-1 and DVL-3protein was mainly expressed in the preputial mesenchymal cell layer. The expression of DVL-1 and DVL-3 protein in foreskin of hypospadias was significantly increased compared with the normal control. Conclusion: The abnormal expression of DVL-1 and DVL-3 is related to the occurrence of hypospadias. The abnormal regulation of DVL-1 and DVL-3 may be one of the main mechanisms leading to hypospadias.
