目的从厚朴浸膏中筛选抑制α-糖苷酶物质,并考察其对糖分吸收的抑制作用。方法建立α-糖苷酶抑制剂的体外筛选模型,追踪厚朴浸膏中α-糖苷酶抑制物质的活性;建立四氧嘧啶所致大鼠糖尿病模型,考察此物质对模型大鼠糖吸收的影响。结果厚朴浸膏的分离样品中厚朴酚的抑制作用最强,其IC50(反应终浓度)=0.16mg·mL-1。结论厚朴酚能够抑制糖尿病大鼠对糖分的吸收,但其作用强度不如拜糖平。
OBJECTIVE To screen inhibitory substances of a-glucosidase from Magnolia officinalis extract and investigate its inhibitory effect on the absorption of sugar. Methods The in vitro screening model of α-glycosidase inhibitor was established to trace the activity of α-glycosidase inhibitory substance in Magnolia officinalis extract. The diabetic model induced by alloxan was established and the effect of this substance on the sugar absorption of model rats . Results The inhibitory effect of magnolol was the strongest in isolated samples of Magnolia officinalis extract with IC50 (final concentration) = 0.16 mg · mL-1. Conclusion Honokiol can inhibit the absorption of sugar in diabetic rats, but its intensity is not as good as that of orally administered sugar.
