[目的]建立并完善石榴叶总黄酮提取和含量测定方法,筛选出最佳提取工艺条件。[方法]以芦丁为对照品,采用分光光度法在最大吸收波长510 nm处对其含量进行测定。[结果]测定石榴叶中的总黄酮含量为0.774%(0.774 mg/g)。最佳提取工艺为乙醇浓度70%、料液比1∶50(g/ml)、超声提取时间为30 min。[结论]选用芦丁为对照品应用于分光光度法测定石榴叶总黄酮含量,方法简单、准确度高、用时短,可用于石榴叶总黄酮的含量测定。
[Objective] The research aimed to establish and perfect the extraction and determination of total flavonoids in pomegranate leaves and screen out the best extraction conditions. [Method] With rutin as reference substance, the content of rutin was determined by spectrophotometry at the maximum absorption wavelength of 510 nm. [Result] The content of total flavones in pomegranate leaf was 0.774% (0.774 mg / g). The best extraction process was ethanol concentration 70%, solid-liquid ratio 1:50 (g / ml), ultrasonic extraction time 30 min. [Conclusion] The application of rutin as reference substance in spectrophotometric determination of total flavonoids in pomegranate leaves was simple, accurate and short in time, and could be used to determine the content of total flavonoids in pomegranate leaves.
