目的探讨信号转导和转录激活因子3(STAT3)活化形式——磷酸化STAT3(p-STAT3)和上皮型钙粘素(E-cadherin)在肝细胞癌中的表达、相互关系及其临床预后意义。方法免疫组化法检测100例临床肝癌标本及10例正常肝组织标本中的p-STAT3和E-cadherin的表达,分析其与肝癌临床病理特征的相关性;以Kap lan-M e ier生存分析及Logrank检验其表达水平与肝癌患者总体生存的关系。结果 p-STAT3在肝癌中呈高表达(58%),与肿瘤侵袭转移密切相关(P=0.038);52%肝癌组织中存在E-cadherin表达降低,与肿瘤侵袭转移亦存在明显相关性(P=0.001);在肝癌组织中p-STAT3与E-cadherin表达呈显著负相关(P=0.006)。生存分析提示p-STAT3阳性表达、E-cadherin降低表达预示肝癌患者不良预后(P=0.005,P=0.001)。结论 STAT3信号通路活化、E-cadherin异常表达与肝癌侵袭转移密切相关,p-STAT3及E-cadherin有望作为肝癌患者预后判断的参考指标。
Objective To investigate the expression and correlation of signal transducer and activator of transcription 3 (STAT3) - phospho-STAT3 (p-STAT3) and E-cadherin in hepatocellular carcinoma significance. Methods The expressions of p-STAT3 and E-cadherin in 100 specimens of hepatocellular carcinoma and 10 specimens of normal liver tissue were detected by immunohistochemistry, and their correlations with the clinicopathological characteristics of hepatocellular carcinoma were analyzed. Kaplan-Meier survival analysis And Logrank test the relationship between the expression level and the overall survival of patients with liver cancer. Results The high expression of p-STAT3 in HCC (58%) was closely related to tumor invasion and metastasis (P = 0.038). The expression of E-cadherin in 52% of HCC tissues was significantly lower than that in invasiveness and metastasis = 0.001). There was a significant negative correlation between the expression of p-STAT3 and E-cadherin in HCC tissues (P = 0.006). Survival analysis indicated that the expression of p-STAT3 was positive, while the decreased expression of E-cadherin predicted the poor prognosis of patients with HCC (P = 0.005, P = 0.001). Conclusion The activation of STAT3 signaling pathway and the abnormal expression of E-cadherin are closely related to the invasion and metastasis of hepatocellular carcinoma. The expressions of p-STAT3 and E-cadherin are expected to be used as prognostic indicators in patients with hepatocellular carcinoma.
