[目的]建立小鼠结肠Cajal间质细胞(ICC)的体外培养方法,探讨大黄素对体外培养结肠ICC的毒-效作用。[方法]酶解消化法体外培养KM乳鼠结肠ICC,运用免疫荧光法鉴定结肠ICC,并用光学显微镜观察其生长期的形态变化。应用均匀设计法和MTT比色法探索大黄素不同浓度和作用时间对结肠ICC的影响。[结果]大黄素对结肠ICC最大毒性浓度为1%,最小毒性浓度为0.001%;最大药效浓度为0.000 1%,最小药效浓度为0.000 008%。[结论]成功建立了体外培养KM乳鼠的结肠ICC的方法。大黄素对结肠ICC既有毒性又有药效作用,毒性浓度范围为0.001%~1%;药效浓度范围为0.000 008%~0.000 1%。
[Objective] To establish an in vitro culture method of colonic Cajal interstitial cells (ICC) in mice and explore the toxic-effect of emodin on ICC of colon in vitro. [Method] Enzyme digestion method was used to culture colonic ICC of KM suckling mice. ICC of colonic mucosa was identified by immunofluorescence method. The morphological changes of colon ICC were observed by light microscopy. The application of uniform design and MTT colorimetric method to explore the different concentrations of emodin and the effect of time on the colon ICC. [Result] The maximum ICC concentration of emodin on colon was 1%, the minimum toxicity concentration was 0.001%, the maximum effective concentration was 0.000 1% and the minimum effective concentration was 0.000 008%. [Conclusion] The method of culturing colonic ICC of KM sucking mice in vitro was successfully established. Emodin on the colon ICC both toxic and pharmacodynamic effects, toxic concentrations ranged from 0.001% to 1%; efficacy range of 0.000 008% to 0.000 1%.
