目的:探讨莪术醇体外抗白血病的作用及机制,为开发利用莪术醇提供理论依据。方法:采用机械分散法结合超声波法制备莪术醇脂质体。以MTT法测定莪术醇等对小鼠淋巴细胞白血病L1210细胞生长的抑制作用。用台盼蓝拒染法及Hoechst 33258/PI双染色,激光共聚焦显微镜观察莪术醇诱导白血病L1210细胞凋亡的作用。结果:莪术醇能显著抑制白血病L1210细胞生长,诱导白血病L1210细胞凋亡。结论:莪术醇诱导肿瘤细胞凋亡是其抗肿瘤作用的重要机制之一。
Objective: To investigate the effect and mechanism of curcumol on leukemia in vitro and to provide a theoretical basis for the development and utilization of curcumol. Methods: The curcumol liposomes were prepared by mechanical dispersion method and ultrasonic method. Inhibitory effect of curcumol on the growth of mouse lymphoblastic leukemia cell line L1210 by MTT assay. Trypan Blue exclusion method and Hoechst 33258 / PI double staining, laser confocal microscopy of curcumol-induced apoptosis in leukemia L1210 cells. Results: Curcumol could significantly inhibit the growth of leukemia L1210 cells and induce the apoptosis of leukemia L1210 cells. Conclusion: Curcumol induced tumor cell apoptosis is one of its important anti-tumor mechanisms.
