目的探讨信号传导和转录激活因子3(STAT3),基质金属蛋白酶-2(MMP-2)和血管内皮生长因子(VEGF)在视网膜母细胞瘤(RB)中的表达及其与临床病程的关系。方法用免疫组织化学SP法分别检测39例RB中p-stat3、MMP-2及VEGF的表达;用全自动医学图像分析系统(LEICA IM,50型)测量其平均灰度值,比较不同临床和病理阶段RB中表达p-STAT3、MMP-2和VEGF的差异及三者阳性表达之间的关系。结果39例RB标本中p-STAT3阳性表达26例,占66.7%(26/39);MMP-2阳性表达31例,占79.5%(31/39);VEGF阳性表达25例,占64.1%(25/39);p-STAT3、MMP-2和VEGF表达水平均与RB视神经浸润呈正相关(P<0.05、P<0.01、P<0.01)与病理分化程度呈负相关(P<0.01、P<0.01、P<0.05),而与患者年龄、性别以及眼别差异均无统计学意义(P>0.05);p-STAT3和MMP-2表达水平呈密切相关(P<0.01),p-STAT3与VEGF表达水平成密切相关(P<0.01)。结论p-STAT3、MMP-2、VEGF的表达水平可能与RB的浸润转移有关,三者共同促进RB的浸润转移。
Objective To investigate the expression of signal transducer and activator of transcription 3 (STAT3), matrix metalloproteinase-2 (MMP-2) and vascular endothelial growth factor (VEGF) in retinoblastoma (RB) and their relationship with clinical course. Methods The expression of p-stat3, MMP-2 and VEGF in 39 cases of RB were detected by immunohistochemical SP method. The average gray value was measured by automatic medical image analysis system (LEICA IM, 50 type) The relationship between the expression of p-STAT3, MMP-2 and VEGF in pathological stage RB and the positive expression of these three. Results The positive expression of p-STAT3 in 39 RB specimens was 66.7% (26/39), the positive expression rate of MMP-2 was 79.5% (31/39), the positive expression of VEGF was 25 (64.1%) 25/39). The expressions of p-STAT3, MMP-2 and VEGF were positively correlated with optic nerve infiltration in RB (P <0.05, P <0.01, P <0.01) 0.01, P <0.05), while there was no significant difference between them (P> 0.05). There was a significant correlation between the expression of p-STAT3 and MMP-2 VEGF expression is closely related (P <0.01). Conclusion The expression of p-STAT3, MMP-2 and VEGF may be related to the infiltration and metastasis of RB, and the three together promote the infiltration and metastasis of RB.
