目的:观察壮肾1号颗粒对阿霉素肾病大鼠蛋白尿模型的外周血CD44、尿β2-MG的影响。方法:用尾静脉一次性注射阿霉素制备肾炎蛋白尿模型,分为5组,分别为空白对照组、模型对照组、洛汀新组、壮肾1号颗粒低剂量组、壮肾1号颗粒高剂量组,观察8周,检测大鼠的尿24h尿蛋白定量、β2-MG,取血流式细胞术检测CD44阳性细胞数。结果:实验8周末各治疗组(壮肾1号颗粒高、低剂量组及洛汀新组)阿霉素肾病大鼠24h尿蛋白定量下降,以壮肾1号颗粒高剂量组最为明显(P<0.05);尿β2-MG的含量降低,以壮肾1号颗粒高剂量组最为明显(P<0.05);阿霉素肾病大鼠CD44阳性细胞数降低,以壮肾1号颗粒高剂量组最为明显(P<0.05)。结论:壮肾1号颗粒能够在一定程度上减少阿霉素肾病大鼠尿蛋白的排泄量,对肾小球有保护作用;减少阿霉素肾病大鼠尿β2-MG的含量,对肾小管有保护作用;降低CD44阳性细胞数降低,提示壮肾1号颗粒通过对阿霉素肾病大鼠外周血CD44阳性细胞数的降低,减轻炎症性损害的发生,影响机体致炎及免疫损伤,从而减少尿蛋白的可能机制。高剂量组对于上述作用较中药低剂量组、洛汀新组疗效较好。
Objective: To observe the effect of Zhuangshen No.1 on peripheral CD44 and urinary β2-MG in proteinuria model of adriamycin-induced nephropathy rats. Methods: Nephritic proteinuria model was established by injecting doxorubicin in the tail vein. The models were divided into 5 groups: blank control group, model control group, Lotensin group, Zhuangshen No.1 granule low dose group, Zhuangshen No.1 In the high-dose group, 8-week urinary proteinuria and β2-MG were detected in urine of rats, and the number of CD44 positive cells was detected by flow cytometry. The content of urinary β2-MG decreased significantly in Zhuangshen No. 1 granule high-dose group (P <0.05). The number of CD44 positive cells in adriamycin-induced nephropathy rats decreased. The most obvious (P <0.05). Conclusion: Zhuangshen 1 granules can reduce urinary protein excretion to a certain extent in adriamycin-induced nephropathy rats and protect glomerulus; decrease urinary β2-MG content in adriamycin-induced nephropathy rats, Reduce the number of CD44-positive cells, suggesting that Zhuangshen 1 particles through the adriamycin nephropathy rat peripheral blood CD44-positive cells to reduce the number of inflammatory damage, the impact of inflammation and immune damage to the body Possible mechanism of reducing urinary protein. High-dose group for the above effect than the traditional Chinese medicine low-dose group, Lotensin better effect.
