目的:研究雄性激素对C57BL/6j雌性小鼠骨髓巨噬细胞凋亡的影响。方法:溴化丙锭染色及透射电镜观察凋亡特征性形态学变化。来自雄性激素和油处理过的小鼠骨髓巨噬细胞分别用杜氏利什曼原虫(Leishmaniadonovani)攻击24h后,检测特征性的DNA凋亡梯形。结果:在培养基中去掉M-CSF后可以诱导骨髓巨噬细胞的凋亡。DNA片段电泳提示:①在雄性激素和油处理组间凋亡细胞的量没有区别,然而②经杜氏利什曼原虫攻击后,雄性激素处理组的凋亡细胞数明显低于油处理组。结论:雄性激素可以抑制感染了杜氏利什曼原虫的巨噬细胞的凋亡,不感染利什曼原虫时,这种抑制作用并不出现。雄性激素对骨髓巨噬细胞凋亡的抑制作用可能在雄性激素诱导的免疫抑制作用中发挥着重要的作用
Objective: To investigate the effect of androgen on the apoptosis of bone marrow macrophage in C57BL / 6j female mice. Methods: The morphological changes of apoptosis were observed by staining with propidium bromide and transmission electron microscopy. Mice bone marrow macrophages from male hormone and oil-treated mice were individually challenged with Leishmaniadonovani for 24 hours to detect characteristic apoptotic DNA trapezias. Results: Apoptosis of bone marrow macrophages can be induced by removing M-CSF in the medium. The electrophoresis of DNA fragments suggested that there was no difference in the amount of apoptotic cells between the male hormone and oil treatment groups, however, ② the number of apoptotic cells in the androgen treatment group was significantly lower than that in the oil treatment group after Leishmania donovani challenge. CONCLUSION: Androgen inhibits the apoptosis of Leishmania donovani-infected macrophages and does not appear when Leishmania is not infected. The inhibitory effect of androgens on the apoptosis of bone marrow macrophages may play an important role in androgen-induced immunosuppression
