目的利用无菌小鼠建立新疆哈萨克族肥胖儿童菌群人源化(HFA)动物模型,研究哈萨克族肥胖儿童的肠道菌群在无菌小鼠体内的定植规律,为深入研究哈萨克族儿童肥胖的病理生理机制提供了一种新的选择方式。方法收集新疆哈萨克族肥胖儿童新鲜粪便样本,选取20只无菌级C57BL/6J小鼠(雌雄各半),于无菌小鼠3周龄时,开始灌胃接种哈萨克族肥胖儿童粪便菌悬液,接种1个月后收集HFA小鼠新鲜粪便样本,利用PCR-DGGE技术进行检测,对DGGE图谱进行相似性UPGMA聚类分析,评价哈萨克族肥胖儿童肠道菌群在HFA小鼠体内的定植规律。结果 HFA小鼠的菌群平均丰富度S为哈萨克族肥胖儿童的0.76倍;HFA小鼠的菌群平均均匀度E及多样性指数H均低于哈萨克族肥胖儿童,但差异无明显统计学意义;HFA小鼠与哈萨克族肥胖儿童肠道菌群的总相似度为64%,而HFA小鼠之间相似度为73%~94%;大部分雌性HFA小鼠与雄性HFA小鼠在聚类分析图上分离。结论 (1)本实验成功利用无菌C57BL/6J小鼠模拟哈萨克族肥胖儿童的特征性肠道菌群环境;(2)该HFA动物模型是探索哈萨克族儿童特征性肥胖与肠道菌群关系的一种新的选择方式。
OBJECTIVE: To establish a humanized (HFA) animal model of obese children in Xinjiang Kazak by using sterile mice to study the colonization rules of intestinal flora of obese children in Kazakstan. In order to further study the obesity of Kazak children, The pathophysiology provides a new way to choose. Methods Fresh stool samples from Kazak obese children in Xinjiang were collected and 20 sterile C57BL / 6J mice (male and female mice) were selected. At the age of 3 weeks, the feces of Kazakh obese children , One month after inoculation, fresh HFA mice stool samples were collected and detected by PCR-DGGE. UPGMA clustering analysis of DGGE profiles was used to evaluate the colonization of intestinal microflora in HFA mice in Kazakh obese children . Results The average abundance of bacteria in HFA mice was 0.76 times of that of obese children in Kazaks. The average evenness E and diversity index H of HFA mice were lower than that of obese children in Kazaks, but the difference was not statistically significant . The overall similarity between HFA mice and Kazakh obese children was 64%, while the similarity between HFA mice was 73% ~ 94%. Most female HFA mice and male HFA mice were clustered Separated from the analysis chart. Conclusions (1) In this experiment, aseptic C57BL / 6J mice were successfully used to simulate the characteristic intestinal microflora environment of Kazak obese children; (2) The HFA animal model was to explore the relationship between Kazakh’s characteristic obesity and intestinal microflora A new way to choose.
