目的:研究自身抗原 -谷氨酸脱羧酶(GAD)预防非肥胖糖尿病(NOD)雌鼠胰岛炎的作用及胰岛炎与NK细胞浸润的相互关系。方法 :应用本室自制猪GAD组 ,腹腔注射于4周龄NOD雌鼠 ,对照组注射等量小牛血清(BSA组) ,分别于19周龄、24周龄和47周龄处死。结果 :同周龄GAD组胰岛炎发生程度和胰岛炎评分均数明显低于同周龄BSA组 ,其评分均数之间存在高度显著性差异(P<0.001) ;同周龄GAD组胰岛内NK细胞计数阳性率明显低于同周龄BSA组 ,有高度显著性差异(P<0.001);胰岛炎分值与胰岛内NK细胞计数阳性率之间存在高度正相关(r=0.7713,P<0.01)。结论 :本室自制的猪GAD能预防NOD雌鼠胰岛炎的发生和抑制NK细胞浸润胰岛内的作用
Objective: To investigate the role of autoantigen - glutamate decarboxylase (GAD) in the prevention of insulitis in nonobese diabetic rats and the relationship between insulitis and NK cell infiltration. Methods: The GAD group was injected intraperitoneally with 4-week-old NOD female rats. The control group was injected with equal amount of bovine serum (BSA group), and were sacrificed at 19 weeks, 24 weeks and 47 weeks respectively. Results: The incidence of insulitis and insulitis in GAD group were significantly lower than those in the same age group (P <0.001). In GAD group, The positive rate of NK cell count was significantly lower than that of the same age BSA group (P <0.001). There was a highly positive correlation between the insulitis score and the positive rate of NK cell count in islet (r = 0.7713, P < 0.01). Conclusion: GAD prepared in our laboratory can prevent the occurrence of insulitis and inhibit the infiltration of NK cells into islets in NOD female mice
