目的:建立测定人体血浆中苦参素的HPLC-MS快速测定方法。方法:血样经乙腈沉淀蛋白后取上清液进样,乙腈: 10 mmol·L-1醋酸铵(含0.08%甲酸)(65:35)为流动相,Nucleodurfe CN柱分离后进入质谱,采用电喷雾电离源正离子模式(ESI+)选择性离子监测(SIR)准分子离子峰。结果:苦参素浓度在12.5-2000 ng·ml-1范围线性良好(r=0.999 1),日内日间RSD均小于5%,回收率大于95%。结论:该法操作简便、快速、准确。
Objective: To establish a rapid HPLC-MS method for the determination of oxymatrine in human plasma. METHODS: The blood sample was precipitated with acetonitrile and the supernatant was sampled. Acetonitrile: 10 mmol·L-1 ammonium acetate (containing 0.08% formic acid) (65:35) was used as the mobile phase. The Nucleodurfe CN column was separated and then passed into mass spectrometry. The electrospray ionization source positive ion mode (ESI+) selective ion monitoring (SIR) excimer ion peak was used. RESULTS: The oxymatrine concentration was linear in the range of 12.5-2000 ng.ml-1 (r=0.999 1). The RSD was less than 5% during the day and the recovery was greater than 95%. Conclusion: The method is simple, rapid and accurate.
