目的 为提高 Prion病诊断率 ,解决 Pr P抗原来源不足 ,人工合成人类 Pr P多肽用于抗体制备。方法 应用固相合成法合成 15个氨基酸残基的人类 Pr P多肽 ,并利用碳二亚胺法将其与牛血清白蛋白偶连制成免疫原 ,应用于动物免疫及多克隆抗体的制备。结果 偶连后 Pr P多肽具有免疫原性 ,并获得了抗 Pr P多克隆抗体。结论 人类 Pr P合成肽抗原的获得和应用 ,可以部分替代天然抗原 ,为进一步单克隆抗体制备 ,以及 Prion病的深入研究提供了有利条件
Aim To improve the diagnosis rate of Prion disease and solve the problem of Pr P antigen source, human Pr P polypeptide was synthesized for antibody preparation. Methods The human Pr P polypeptide with 15 amino acid residues was synthesized by solid phase synthesis. The conjugate was made into bovine serum albumin conjugate by carbodiimide method, which was used in animal immunization and polyclonal antibody preparation. As a result, the PrP polypeptide was immunogenic after conjugation, and a polyclonal anti-PrP antibody was obtained. Conclusion The obtainment and application of human Pr P synthetic peptide antigen can partly replace natural antigen, which provides favorable conditions for the further preparation of monoclonal antibody and in-depth study of Prion disease
