目的:观察氯沙坦对肝星状细胞增殖和凋亡的影响。方法:采用CCK8试剂盒检测氯沙坦对肝星状细胞增殖的抑制情况;重复实验可计算IC50;采用AnnexinV-FITC/PI双染法经流式细胞仪检测肝星状细胞的凋亡情况。结果:氯沙坦可抑制肝星状细胞的增殖,并呈明显的剂量依赖性,1000μmol·L-1时肝细胞的抑制率可达(95.22±3.54)%,IC50值为(326.21±30.27)μmol·L-1;AnnexinV-FITC/PI双染法检测显示,各浓度的氯沙坦均能促进细胞凋亡,与对照组比较有显著性差异(P<0.01)。结论:氯沙坦有抑制肝星状细胞细胞增殖及促进其凋亡的作用。
Objective: To observe the effects of losartan on the proliferation and apoptosis of hepatic stellate cells. Methods: The inhibitory effect of losartan on the proliferation of hepatic stellate cells was detected by CCK8 kit. The IC50 was calculated by repeated experiments. The apoptosis of hepatic stellate cells was detected by flow cytometry with Annexin V-FITC / PI double staining. RESULTS: Losartan inhibited the proliferation of hepatic stellate cells in a dose-dependent manner. The inhibitory rate of hepatocytes was (95.22 ± 3.54)% with IC50 value of (326.21 ± 30.27) at 1000 μmol·L-1, The results of Annexin V-FITC / PI double staining showed that all the concentrations of losartan could promote apoptosis, which was significantly different from the control group (P <0.01). Conclusion: Losartan can inhibit hepatic stellate cell proliferation and promote apoptosis.
