目的:观察加味六君子汤含药血清对胃癌BGC-823细胞增殖和凋亡的影响。方法:以加味六君子汤不同浓度含药血清培养BGC-823细胞后,以MTT法检测对细胞增殖的影响,流式细胞仪检测细胞周期和细胞凋亡变化。结果:浓度为10%、15%、20%、30%、40%的含药血清作用于BGC-823细胞24、48、72小时后,细胞存活率显著降低,且与时间和剂量呈依赖关系。浓度为10%、15%、20%的含药血清作用于BGC-823细胞24小时后,G2期细胞增多,S期细胞减少。加药组BGC-823细胞凋亡率分别为(13.27±0.88)%、(19.97±0.60)%和(33.09±0.58)%,与空白对照组相同浓度空白血清细胞凋亡率(0.34±0.02)%、(0.19±0.04)%和(0.18±0.03)%比较,差异均有统计学意义(P<0.01)。结论:加味六君子汤含药血清对BGC-823细胞生长有明显抑制作用,并可诱导BGC-823细胞凋亡。
Objective: To observe the effects of Jiawei Liujunzi Decoction serum on proliferation and apoptosis of gastric cancer cell line BGC-823. Methods: BGC-823 cells were cultured in different concentrations of Jiajun Liujunzi Decoction. Cell proliferation was detected by MTT assay. Cell cycle and apoptosis were detected by flow cytometry. Results: After treated with 10%, 15%, 20%, 30% and 40% serum for 24,48,72 hours, BGC-823 cells significantly decreased cell viability and time-and dose-dependent . Concentration of 10%, 15%, 20% of the serum containing BGC-823 cells for 24 hours after the G2 phase cells increased, S phase cells decreased. The apoptotic rates of BGC-823 cells in drug-treated group were (13.27 ± 0.88)%, (19.97 ± 0.60)% and (33.09 ± 0.58)%, respectively. Compared with blank control group, the apoptosis rate of blank serum was (0.34 ± 0.02) %, (0.19 ± 0.04)% and (0.18 ± 0.03)%, respectively. The difference was statistically significant (P <0.01). Conclusion: Modified Liujunzi decoction serum can significantly inhibit the growth of BGC-823 cells and induce the apoptosis of BGC-823 cells.
