目的建立检测血清中十八碳三烯酸(ALA)、二十碳五烯酸(EPA)2种多不饱和脂肪酸的新方法,并用该方法检测危重症患者与正常人血清中ALA、EPA的含量。方法将人血清中的十八碳三烯酸(ALA)、二十碳五烯酸(EPA)经正己烷萃取浓缩,经硫酸-甲醇甲酯化,用气相色谱法定性、定量检测。结果危重患者和正常人血清中的ALA、EPA含量差异均无统计学意义(P>0.05)。检出限(S/N=3):ALA:2.3μg/ml,EPA:1.4μg/ml。ALA,EPA的RSD(n=6)分别为2.32%、3.94%。结论此方法重复性好、敏感度高,可用于Omega-3多不饱和脂肪酸含量的测定。
Objective To establish a new method for the determination of two polyunsaturated fatty acids, octadecatrienoic acid (ALA) and eicosapentaenoic acid (EPA), in sera of critically ill patients and normal subjects, and to detect the levels of ALA and EPA content. Methods The octadecatrienoic acid (ALA) and eicosapentaenoic acid (EPA) in human serum were extracted by n-hexane and concentrated by sulfuric acid-methanol methyl ester. Qualitative and quantitative detection by gas chromatography. Results Serum levels of ALA and EPA in critically ill patients and normal persons showed no significant difference (P> 0.05). Detection limit (S / N = 3): ALA: 2.3 μg / ml, EPA: 1.4 μg / ml. The RSD (n = 6) of ALA and EPA were 2.32% and 3.94% respectively. Conclusion This method has good repeatability and high sensitivity and can be used to determine the content of polyunsaturated fatty acids in Omega - 3.
