目的研究植酸(IP6)对人胶质母细胞U118MG生长及增殖的影响并探讨其作用机制。方法体外培养U118MG细胞,使用MTT实验法检测植酸对U118MG细胞的增殖抑制影响;Western blot检测Bcl-2、Caspase-3、p53的蛋白水平。结果 MTT实验表明植酸具有抑制U118MG细胞生长的作用(P<0.05),并与剂量、时间呈正关系;植酸可抑制Bcl-2蛋白表达,且诱导Caspase-3、p53的活化,具有统计学差异(P<0.05)。结论植酸具有抑制U118MG细胞增殖、诱导凋亡的作用,Bcl-2、Caspase-3、p53可能参与植酸对人胶质母细胞U118MG的诱导凋亡机制。
Objective To study the effect of phytic acid (IP6) on the growth and proliferation of human glial cell line U118MG and to explore its mechanism. Methods U118MG cells were cultured in vitro. The effects of phytic acid on the proliferation of U118MG cells were detected by MTT assay. The protein levels of Bcl-2, Caspase-3 and p53 were detected by Western blot. Results MTT assay showed that phytic acid could inhibit the growth of U118MG cells (P <0.05), and had a positive correlation with dose and time. Phytic acid could inhibit the expression of Bcl-2 and induce the activation of Caspase-3 and p53 with statistical significance Difference (P <0.05). Conclusion Phytic acid can inhibit the proliferation and induce apoptosis of U118MG cells. Bcl-2, Caspase-3 and p53 may be involved in the mechanism of phytic acid inducing apoptosis of human glial cell line U118MG.
